Uncovering key flavor-related pathways in mango cultivars from Yunnan by integrated transcriptomics and metabolomics analyses

摘要: Mango flavor is crucial for sensory perception and varies significantly among cultivars, yet its gene regulatory mechanisms remain unclear. This study investigated flavor differences and biosynthetic mechanisms among 5 mango cultivars grown in Yunnan's dry-hot valley climate, utilizing integrated transcriptomics and metabolomics approaches. Three sugars (glucose, fructose, sucrose), 11 organic acids, 16 free amino acids, and 80 volatile compounds were identified. Among them, sucrose, citric acid, and cysteine were the predominant sugar, organic acid and free amino acid, respectively. Terpenes, particularly beta-ocimene, 3-carene and terpinolene, were the dominant volatile compounds. Kyoto Encyclopedia of Genes and Genomes pathway analysis revealed 20, 60, and 74 structural genes involved in sugars, organic acids and volatiles metabolism, respectively. Weighted gene co-expression network analysis (WGCNA) identified 51 key genes across 5 modules, including SPS, ADH, PEPC, CS and DXS. Eight transcription factors (TFs), such as bZIP60, TGA7, WRKY57, bHLH143, GATA19, MYB1, ERF118 and bZIP44, were implicated in flavor metabolism. Glucose, malic acid and methyl heptanone emerged as key metabolites, significantly associated with 23, 16 and 17 differentially expressed genes, respectively. This study constructed a comprehensively regulatory network of mango flavor metabolism, providing insights into mango flavor development in Yunnan cultivars, supporting breeding and cultivation improvements.
