Identifying QTLs for the Anthocyanin Composition of Pigmented Potato Flesh in a Diploid Potato Population

摘要: Red- and purple-fleshed potatoes contain anthocyanins that are beneficial for health, leading to high consumer demand. The identification of favorable alleles for use in potato breeding may help satisfy this commercial demand. In this study, to identify novel QTLs associated with anthocyanin content in potato tubers, a hybrid population of 208 F1 offspring was established by crossing red-fleshed diploid maternal parents and purple-fleshed diploid paternal parents. The tuber flesh of individuals in the resulting population was analyzed for six main anthocyanins, delphinidin (Dp), cyanidin (Cy), petunidin (Pt), pelargonidin (Pg), peonidin (Pe), and malvidin (Mv), using high-performance liquid chromatography (HPLC). High-quality SNP markers were obtained through restriction site-associated DNA (RAD) tag sequencing technology, with 9556 markers selected for further analysis. A high-density map was constructed for quantitative trait locus (QTL) mapping of the six anthocyanins, resulting in the discovery of a novel QTL cluster including 8 QTLs. This QTL cluster was located in the 40.07-46.68 Mb region of chromosome 10. These findings offer new insight into the formation of anthocyanins in tuber flesh and provide a foundation for the fine mapping and cloning of genes that regulate the anthocyanin composition of tubers.
