Amelioration of Kluyveromyces marxianus GY-8 on high-fat diet-induced obesity and gut microbiota dysbiosis

摘要: The rising prevalence of diet-induced obesity underscores the need for novel interventions targeting metabolic dysregulation and gut microbiota imbalance. The aim of this study was to evaluate the intervention effect of the potential probiotic Kluyveromyces marxianus GY-8 isolated from Tibetan kefir on obesity. This strain demonstrated notable lipid-lowering ability in vitro, exhibiting a cholesterol assimilation rate of 50.97 % and triglyceride degradation rate of 42.30 %. Further, K. marxianus GY-8 was administered orally to high-fat diet-induced obese mice to assess its intervention effects. The results suggested that K. marxianus GY-8 treatment significantly reduced body weight, improved the hepatic lipid accumulation and decreased liver damage by reducing heptic triglyceride (TG), total cholesterol (TC), and malondialdehyde (MDA) levels. Moreover, K. marxianus GY-8 treatment downregulated expression of carnitine palmitoyl transferase-1 (CPT1), sterol regulatory element binding protein-1c (SREBP-1c), and acetyl-CoA carboxylases alpha (ACACA), thereby improving hepatic lipid metabolism. In addition, K. marxianus GY-8 treatment modulated the dysbiotic gut microbiota, by enriching taxa associated with short-chain fatty acids (SCFAs) production, particularly propionic and butyric acid, which correlated with ameliorated metabolic dysfunction. Therefore, it can be concluded that K. marxianus GY-8 alleviates obesity through the dual mechanisms of regulating heptic lipid metabolism and restoration of gut microbiota balance. These findings may provide theoretical basis for the application of K. marxianus GY-8 in the development of anti-obesity related functional foods.
